學(xué)術(shù)與應(yīng)用研究:

從事天然產(chǎn)物心血管藥理學(xué)活性與機(jī)制研究。重點(diǎn)方向是具有心血管藥理活性并具有重復(fù)結(jié)構(gòu)單元的天然產(chǎn)物多糖的化學(xué)結(jié)構(gòu),、結(jié)構(gòu)修飾,、藥效活性、藥理機(jī)制及構(gòu)效關(guān)系研究,。

巖藻糖化糖胺聚糖(fucosylated glycosaminoglycan, FG)結(jié)構(gòu)研究方面,,基于“bottom up”策略,,通過(guò)建立糖苷鍵選擇性解聚技術(shù)及寡糖純化與波譜學(xué)結(jié)構(gòu)解析技術(shù),闡明了一系列不同物種來(lái)源的天然FG的確切結(jié)構(gòu),;獲得一系列用于藥理學(xué)活性與機(jī)制研究的,、具有特殊端基結(jié)構(gòu)的純品FG寡糖及其結(jié)構(gòu)修飾產(chǎn)物。

系統(tǒng)研究了FG及其解聚產(chǎn)物,、純品寡糖的抗凝抗血栓活性及其藥理學(xué)機(jī)制特點(diǎn),,重點(diǎn)研究了FG寡糖選擇性抑制內(nèi)源性因子X(jué)酶(Tenase, Xase)構(gòu)效關(guān)系、必須結(jié)構(gòu),、酶學(xué)動(dòng)力學(xué)特點(diǎn)與機(jī)制,、靶蛋白相互作用的動(dòng)力學(xué)過(guò)程與機(jī)制。

基于天然FG結(jié)構(gòu)修飾,,研制選擇性抑制內(nèi)源性Xase的新靶點(diǎn)抗凝抗血栓新藥LFG-53(First In Class,,獲FDA和NMPA臨床許可),系列專(zhuān)利獲得國(guó)內(nèi),、美,、日等國(guó)授權(quán),已實(shí)現(xiàn)知識(shí)產(chǎn)權(quán)及關(guān)鍵技術(shù)的企業(yè)技術(shù)轉(zhuǎn)移,。

海王集團(tuán)國(guó)家級(jí)企業(yè)技術(shù)中心工作期間,,組建藥物研發(fā)實(shí)驗(yàn)研究平臺(tái),主管創(chuàng)新藥物研發(fā),。其代表性工作包括:立項(xiàng)并研發(fā)中藥天然藥物1類(lèi)新藥虎杖苷注射液(臨床批件(現(xiàn)已取得FDA臨床許可)),、5類(lèi)中藥新藥艾心酮片(臨床批件)、中藥大品種改進(jìn)藥物鼻淵軟膠囊(上市許可)等,。
